The , a world leader in genetic research, is looking for exceptional candidates to join the Cardiovascular Disease Initiative (CVDi). The successful candidate will join a multidisciplinary team to assist in the investigation of cardiovascular disease using genetics, cell biology, biochemistry, cell lines, and model systems. The current projects will leverage recent genetic discoveries to validate potential new targets for cardiovascular diseases under the direction of Patrick Ellinor. The candidate will be involved in day-to-day laboratory work working with research scientists, computational scientists, postdoctoral fellows and other research associates, assisting with experimental design, analyzing and troubleshooting experiments, and communicating scientific results to the Broad CVDi team.

The primary responsibility for this position will be to contribute to the production of iPSC/ES cell derived cardiomyocytes, the development of various cell based assays for target validation, and to support the team at the early stages of drug development. The candidate will conduct experimental research studies at the bench under the supervision of a research scientist.

Key Responsibilities:

  • Maintain/organize of cell lines and cell banks, and isolate primary cells for use in cell-based assays.

  • Culture and differentiate human pluripotent stem cells to cardiomyocytes (hES and hiPSC) for further screens and target validation.

  • Develop and execute cell-based assays to support target validation and hit identification & optimization.

  • Contribute to target validation using genomic perturbation techniques such as CRISPR, and siRNA.

  • Independently perform molecular assays such as PCR, western blots, immunofluorescence and virus preparations for target validation purposes.

  • Design and execute research plans to ensure that critical project requirements, timelines and deliverables are met.

  • Troubleshoot scientific problems with minimal guidance.

  • Analyze experimental data and communicate scientific results to team members.

  • Prepare, order, and maintain stocks of necessary reagents and supplies.

  • Attend project meetings to present results and coordinate with other scientists to support project goals.

  • Contribute to lab manager responsibilities as needed.

Requirements:

  • Bachelor's degree with 3+ years or MS degree with 2+ years of relevant laboratory experience.

  • Experience with molecular biology (cloning, PCR, RT-PCR, CRISPR/Cas-9 technology)

    • Experience with primer design and sequencing technologies/methods.

    • DNA/RNA/protein extraction/purification.

  • Experience with mammalian cell culture and primary cell transfection.

  • Experience with pluripotent stem cell culture, e.g. culturing and differentiating hES and hiPS cells to various cell types is preferred.

  • Hands-on experience in cellular assay development and proficiency with standard assay formats (e.g. flow cytometry, imaging, luminescence-/fluorescence-based cellular readouts, immunohistochemistry) in primary and cultured cells is preferred.

    • High-throughput microscopy setup, Opera Phenix/Harmony software experience preferred.

    • Experience with automated pipetting systems preferred.

  • Ability to critically read, interpret, and adapt scientific literature.

  • Strong communication and presentation skills (verbal and written).

  • Strong organization, documentation and time management skills.

  • Ability to build excellent working relationships and foster collaboration in a team environment.

  • This position entails working on some weekends.
